MACSPI enables tissue-selective proteomic and interactomic analyses in multicellular organisms

ABSTRACT
Multicellular organisms are composed of many tissue types that have distinct morphologies and functions, which largely driven by specialized proteomes interactomes. To define the proteome interactome a specific type in an intact animal, we developed localized proteomics approach called Methionine Analog-based Cell-Specific Proteomics Interactomics (MACSPI). This method uses tissue-specific expression engineered methionyl-tRNA synthetase to label proteins with bifunctional amino acid 2-amino-5-diazirinylnonynoic selected cells. We applied MACSPI Caenorhabditis elegans, model multicellular organism, selectively label, capture, profile body wall muscle nervous system, led identification proteins. Using photo-cross-linker, successfully profiled HSP90 interactors muscles neurons identified stress-related interactors. Our study demonstrates can be used interactomes organisms.
