ABSTRACT
In hematopoiesis, co-expression of Sca-1 and c-Kit defines cells (LS(+)K) with long term reconstituting potential. contrast, poorly characterized LS(-)K fail to reconstitute lethally irradiated recipients. Relative quantification mass spectrometry transcriptional profiling were used characterize LS(+)K cells. This approach yielded data on >1200 proteins. Only 32% protein changes correlated mRNA modulation demonstrating post-translational regulation in early hematopoietic development. had lower expression synthesis proteins but did express associated mature cell function. Major increases erythroid development observed cells; based this assessment potential we showed them be principally progenitors, effective use discovery proteomics for definition primitive
