ABSTRACT
The anaphylatoxin C5a and its receptor C5aR (CD88) are complement pathway effectors implicated in renal diseases, including ANCA-associated vasculitis. We investigated the kidney expression of a second C5L2 by using immunohistochemistry, situ hybridization, spatial gene on formalin-fixed, paraffin-embedded human mouse kidney. was detected interstitial macrophages multiple tubular regions, distal proximal; had similar pattern. 5/6 nephrectomy model chronic injury exhibited increased infiltrating cells within fibrotic regions. confirmed leukocytes vitro differentiated flow cytometry, treatment with induced chemokines remodeling factors macrophages, CCL-3/-4/-7, -20, MMP-1/-3/-8/-12, F3, promoted leukocyte survival. activity dependent, as demonstrated reversal inhibitor avacopan. Collectively, these results suggest that myeloid may induce excessive inflammation via immune cell recruitment, extracellular matrix destruction, remodeling, resulting tissue deposition.
