ABSTRACT
Cdc42 is a small GTPase involved in the regulation of cytoskeleton and cell polarity. To test whether has an essential role formation filopodia or directed migration, we generated Cdc42-deficient fibroblastoid cells by conditional gene inactivation. We report here that loss did not affect filopodium lamellipodium had no significant influence on speed migration nor mitosis. displayed more elongated shape reduced area. Furthermore, directionality during reorientation Golgi apparatus into direction was decreased. However, expression dominant negative Cdc42-null resulted strongly severely single directionality, complete polarization protrusion toward wound, as well membrane blebbing. Thus, our data show besides additional GTPases Rho-family, which share GEFs with Cdc42, are establishment maintenance polarity migration.
