ABSTRACT
Abstract Oculomotor tasks generate a potential wealth of behavioural biomarkers for neurodegenerative diseases. Overlap between oculomotor and disease-impaired circuitry reveals the location severity disease processes via saccade parameters measured from eye movement such as prosaccade antisaccade. Existing studies typically examine few in single diseases, using multiple separate neuropsychological test scores to relate behaviour cognition; however, this approach produces inconsistent, ungeneralizable results fails consider cognitive heterogeneity these Comprehensive assessment direct inter-disease comparison are crucial accurately reveal biomarkers. We remediate issues by characterizing 12 parameters, selected robustly describe behaviour, derived an interleaved antisaccade task large cross-sectional data set comprising five cohorts (Alzheimer’s disease/mild impairment, amyotrophic lateral sclerosis, frontotemporal dementia, Parkinson’s disease, cerebrovascular disease; n = 391, age 40–87) healthy controls (n 149, 42–87). These participants additionally completed extensive battery. further subdivided each cohort diagnostic subgroup (for Alzheimer’s impairment dementia) or degree based on testing (all other cohorts). sought understand links their relationships robust measures, alterations disease. performed factor analysis evaluating interrelationships among examined correlations four resultant factors neuropsychology-based domain scores. then compared abovementioned subgroups at individual parameter level. theorized that underlying integrity distinct task-relevant brain process. Notably, Factor 3 (voluntary generation) 1 (task disengagements) significantly correlated with attention/working memory executive function also visuospatial 2 (pre-emptive global inhibition) only scores, 4 (saccade metrics) no Impairment several mostly antisaccade-related scaled across cohorts, while differed parameters. The detects subsets likely index disparate related different domains. This suggests represents sensitive paradigm can simultaneously evaluate variety clinically relevant constructs diseases could be developed into screening tool applicable diagnoses.
