ABSTRACT
Abstract Actual loss of lizard biodiversity continues, even with the implementation conventional conservation programs. An approach including assisted reproductive techniques such as sperm cryopreservation may contribute to management endangered species. We developed a method for in sceloporine lizards and compared response among studied Prior mating season, we obtained semen from adult males Sceloporus aeneus (n = 21), grammicus 20) torquatus 21) via pressure genital papilla. Volume concentration were measured before dilution Tris–egg yolk (TEY) medium evaluate progressive motility, viability, morphology, plasma membrane acrosome integrity. Then, cooled remaining volumes 5°C at rate 0.1°C per minute incorporate glycerol (8% v/v) two fractions. Immediately afterwards, placed 40 μl mix on solid CO2 form pellets immersed them liquid nitrogen storage. thawed 29°C 3 minutes diluted 1:1 (v/v) TEY assess quality. found positive relationship between body weight seminal volume S. negative correlation (P < 0.05). Moreover, observed that freezing–thawing process decreased quality three species, mostly affecting motility viability. However, showed higher tolerance than grammicus.
