ABSTRACT
MADS-box transcription factor genes are well known for their role in floral organ and seed development. In this study, a novel MADS-box-containing gene, designated NbMADS1, was isolated from leaves of Nicotiana benthamiana. The full-length cDNA 666bp encoded putative polypeptide 221 aa with mass 24.3kDa. To assess the NbMADS1 defence response to bacterial harpinXoo, an elicitor hypersensitive response, loss-of-function experiment performed N. benthamiana plants using virus-induced gene silencing. Analyses electrolyte leakage revealed more extensive cell death control than NbMADS1-silenced plants. showed impaired harpinXoo-induced stomatal closure, decreased production hydrogen peroxide (H2O2) nitric oxide (NO) guard cells, reduced resistance Phytophthora nicotianae. compromised closure observed inhibited by application H2O2 sodium nitroprusside (an NO donor). Taken together, these results demonstrate that NbMADS1–H2O2–NO pathway mediates multiple harpinXoo-triggered responses, including death, defence-related expression, suggesting plays important regulating harpinXoo
