ABSTRACT
Background: MicroRNAs (miRNAs) are small non-coding RNA molecules regulating mRNAs translation. Significant differences in miRNAs expression have been found glioblastoma compared to normal brain tissue. MiR-124a, miR-381 and miR-637 involved regulation of pathways which commonly deregulated (MAPK/ERK, WNT/β-catenin AKT pathways, respectively). AIM: To investigate the profile prognostic/predictive role miR-124a, primary IDH-wildtype glioblastoma. Materials Methods: Inclusion criteria were: pathological diagnosis (according WHO 2016 criteria), partial/total surgical resection followed by at least one line postoperative adjuvant treatment, ≥ 6-months follow-up after diagnosis, Caucasian ethnicity. Histological revision was performed independently two pathologists confirm diagnosis. IDH1/IDH2 status assessed Sanger sequencing according standard protocols. Total extracted using miRNeasy FFPE (QIAGEN), were amplified with TaqMan MicroRNA Assays (ThermoFisher) RNU6B used as endogenous control, while ten areas selected external controls. Relative quantification miRNA values (RQ) calculated ΔΔCt method. X-tile plots Stata (StataCorp LP) calculate RQ cut-off for statistical analyses, respectively. Informed consent obtained local regulations. Results: Here we present preliminary results based on 49 patients, 30 males (61%). Median age 59 years (range 38–75), median KPS before surgery 80 70–90). Partial achieved 27/49 (55%), total 22/49 (45%). progression-free survival (PFS) 9.4 months 3.5–26), overall (OS) 16 3.5–42.7). treatment Stupp regimen both associated a better outcome expected. Glioblastoma samples showed reduced all three brain. Significantly PFS (9.9 vs 6.0 months, p=0.002) patients up-regulated (RQ cut-off: 0.248), whereas OS difference not significant (16.9 15.6, p=0.161). A specific range 0.104–0.287) (9.8 7.4, p=0.0183), but (16.4 15.8, p=0.782). No associations between miR-124a outcome. Conclusions: significantly up-regulation, finding consistent its reported inhibitory effect pathway. Interestingly, favorable seems be limited comprising value. These findings need larger series study is ongoing.
