ABSTRACT
Vigna marina (Burm.) Merr., the dune bean or notched cowpea, is a tropical creeping vine that grows on sand dunes along coastal regions of Taiwan. Although V. weed, some varieties are also grown for fodder and food. This legume natural host Bean common mosaic virus in Solomon Islands (1) Alfalfa Beet western yellows Australia (2). In April 2009, plants showing severe chlorotic ringspots foliage were found region Hualien County eastern Indirect ELISA single diseased plant showed positive results with antibodies against cucumber isolate Cucumber (CMV) but negative to Broad wilt virus-1, virus-2, potyviruses (Agdia Inc., Elkhart, IN). A pure CMV was obtained from through three successive passages lesion isolation sap-inoculated Chenopodium quinoa. Results mechanical inoculations CMV-V. successfully transmitted C. amaranticolor, murale, quinoa, Chrysanthemum coronarium, Gomphrena globosa, Nicotiana benthamiana, N. tabacum cv. Vam-Hicks, Phaseolus limensis, P. lunatus, vulgaris, Tetragonia tetragonioides, marina, radiata, unguiculata subsp. sesquipedalis. These artificial confirmed by ELISA. Homologous reactions stock polyclonal antiserum CMV-cucumber (4) observed sodium dodecyl sulfate-immunodiffusion. To determine specific subgroup, total RNA extracted inoculated leaves quinoa using Total Plant Extraction Miniprep System (Viogene, Sunnyvale, CA). DNA fragment 940 bp covering 3′ end coat protein gene C-terminal noncoding RNA-3 amplified Cucumovirus-specific primers (3) after reverse transcription (RT)-PCR AccuPower RT/PCR PreMix Kit (Bioneer, Daejeon, Korea). The product gel purified Micro-Elute DNA/Clean (GeneMark Technology Co., Tainan, Taiwan) cloned yT&A Cloning Vector (Yeastern Biotech Taipei, sequencing (Mission sequence submitted GenBank (No. HM015286). Pairwise comparisons corresponding sequences other isolates revealed maximum (95 96%) nucleotide identities subgroup IB (strains Nt9 Tfn) compared 94 95% IA Y Fny) 77 78% II LS Q). suggest causal agent disease Taiwan belongs I. our knowledge, this first report as CMV. strain pathogenicity could threaten crop production zones. addition, associated native vegetation injured infection, which might lead ecological impacts shoreline fading.
