First Report of Chickpea Chlorotic Dwarf Virus Infecting Spring Chickpea in Syria

ABSTRACT
During May 2003, a high incidence of symptoms suggestive virus infection in spring chickpea were observed many fields Al-Ghab Valley, Syria, the ICARDA farm (near Aleppo, Syria), as well other locations northern including Idleb governorate. Symptoms yellowing, stunting, and necrosis. A total 1,345 samples with these (331 from 269 farm, 745 governorate) collected tested for presence five viruses tissue-blot immunoassay (TBIA) (4) at Virology Laboratory ICARDA, using following antisera: monoclonal antibodies Faba bean necrotic yellows (FBNYV, genus Nanovirus) (1); Bean leafroll (BLRV, family Luteoviridae) (4B10) (3); Beet western (BWYV, Polerovirus, Luteoviridae [ATCC PVAS-647, American Type Culture Collection, Manassas, VA]); Soybean dwarf (SbDV, Luteoviridae, PVAS-650]) polyclonal Chickpea chlorotic (CpCDV, Mastrevirus, Geminiviridae, provided by H. J. Vetten, BBA, Braunschweig, Germany). The most common present was BWYV (detected 54.1% tested), followed CpCDV (19.2%), BLRV (10.2%), FBNYV (5.5%). SbDV not detected any tested. Using immunosorbent electron microscopy, infected revealed low numbers geminivirus-like particles after 15 min incubation on antiserum-coated grids. When purified plants, coat protein 32 kDa sodium dodecyl sulfate-polyacrylamide gel electrophoresis typical (2) reacted strongly antiserum blots. vector Syria found to be Orosius albicinctus Distant, is thought similar orientalis (Matsumura), reported (2).
