ABSTRACT
NFAT family of transcription factors encodes 5 members and is present in a wide range cell types, regulating genes involved proliferation, angiogenesis, cycle progression apoptosis. has two well conserved regions: the regulatory domain DNA binding domain. N C‐terminal ends are transactivation sites show less sequence similarity. To understand molecular mechanisms related to functions we performed yeast hybrid screening using NFAT1 end as bait. We identified that interacts with IRF2BP2 transcriptional repressor by its RING This interaction seems be specific, since other regions do not interact IRF2BP2. Furthermore, co‐localize nucleus activated cells luciferase report assays demonstrate represses NFAT1‐dependent activation IL‐2 IL‐4 promoters TNF‐alfa k3 element. also nuclear localization signal mutation this abolishes repression function Our data demonstrated repress activity, suggesting can new regulator activity.
