ABSTRACT
Introduction: The indication for liver transplantation (LTx) in case of hepatocellular carcinoma (HCC) is still a sensitive issue since the validation Milan Criteria has been objectively criticized numerous studies. microRNA (miRNA) are currently widely investigated as biomarkers establishment personalized medicine and oncology. miRNA have already introduced diagnosis graft rejection or tolerance cancer diagnosis; however published profiles HCC discordant couldn't verified prospective controlled trial patients. Here we introduce first results from our multicenter database development miRNA-based biomarker monitoring prediction recurrence after LTx. Methods: Since May 2011, total number 105 patients with chronic disease (CL) without included into study centers (Charité - Universitätsmedizin Berlin University Clinic Leipzig). Samples 28 (15 13 HCC) used preliminary analyses. 19 suffered alcoholic (AL) 9 hepatitis C infection (HCV). Total RNA was isolated tissue samples using Trizol reagent measured Nanodrop. miRNA-profiling performed miRCURY LNA platform by exiqon (miRBase version 17). Results: microarray assays showed good quality high expression specific miR-122. miR-3182, miR-491-3p, miR-122, pre-miR122, miR-200a miR-200b were down-regulated whereas miR-4288 up-regulated overall tumor comparing to surrounding tumors. miRNAs pre-let-7b, pre-let7f-1, miR-1247 pre-miR129 expressed at higher levels CL compared cirrhotic In these samples, miR-1207-3p, miR-1249, miR-1250, miR-1279 miR-1281 down-regulated. Although there no difference between HCV patients, 18 significantly different non-tumor AL. Moreover some an up-regulation tendency AL, namely miR-143, miR4328, miR-3613-3p, miR-24, miR-145. Conclusion: one single could affect post-transcriptional regulation up hundred genes, patterns more likely be suitable than levels. Our data shows clear differences miRNA-profiles both Interestingly, signature non-cancerous tissue, which possibly malignancy. signatures differentiate their underlying like AL vs. therefore they serve medicine.
