ABSTRACT
Peripheral T-cell lymphomas (PTCLs) in western countries are uncommon tumors with unfavorable prognosis. They may be subclassified as anaplastic large-cell (ALCLs), angioimmunoblastic-T-cell (AITLs), or unspecified peripheral (PTCLs-U). It has recently been demonstrated that AITLs originate from germinal center follicular helper T cells (TFH), whereas the normal counterparts of other PTCLs remain essentially unknown. The aim this study was to establish whether PTCL subgroups also express TFH cell markers. One hundred forty-six were analyzed for programmed death-1 (PD-1) expression tissue microarrays using a new monoclonal antibody called NAT-105. PD-1-positive cases, which did not fulfill all criteria AITL, further evaluated whole-tissue sections another 12 immunohistochemical markers, including markers CXCL13, CD10, and BCL6. Clonal Ig receptor rearrangements Epstein-Barr virus-encoded RNA evaluated. Morphologic, clinical, follow-up data reviewed.Twenty-five out 87 non-AITL cases (28.75%) showed PD-1 immunostaining. BCL6, CD10 found 24/25 (96%), 16/25 (64%), 6/25 (24%) respectively. All expressed at least 2 markers. Moreover, 5 positive 4 Most (17/25, 68%) displayed some AITL-like features. Of remainder, 1 considered early diagnosed ALCL-anaplastic lymphoma kinase-negative, 6 PTCLs-U had morphology consistent lymphoepithelioid (Lennert's) lymphoma. Three IgH clonal rearrangement, associated virus expression. Our series patients differ significantly their clinical presentation most reported literature: 55% them alive 35% complete remission after median 15 months cyclophosphamide, dexorubicin, vincristine, prednisone-based chemotherapy.TFH especially PD-1, subset classified although shared morphologic features AITL. This suggests spectrum AITL wider than previously thought, possibly Additionally, results suggest subgroup PTCLs-U, distinct denominated ALCL, derived cells, they develop along pathogenic pathway.
