Anti–IL-6 Antibody Treatment but Not IL-6 Knockout Improves Intestinal Barrier Function and Reduces Inflammation After Binge Ethanol Exposure and Burn Injury

ABSTRACT
Interleukin 6 (IL-6) is an inflammatory cytokine known to be elevated in chronic diseases and after insults such as trauma infection. Although necessary for the development of B cells Th17 cells, IL-6, at levels, can also cause tissue damage lead a rise inflammation. Previous work our laboratory has shown that IL-6 increased both systemically multiple organ systems including ileum ethanol exposure burn injury. As this combined insult causes intestinal morphological damage, tight junction protein localization alterations, phosphorylated myosin light chain we sought determine role these responses using model binge antibody treatment reduced changes ileum, bacterial translocation, levels relative either injury alone. Zonula occludens 1 occludin was reestablished wild-type mice given burn. 6-knockout had damage; however, no translocation or were observed compared with similarly treated mice. These data suggest may have injury, although complete loss does not seem beneficial model. Modulation present new option preventing associated inflammation
