ABSTRACT
Five isolates of Cucumber mosaic virus (CMV) from Lilium sp. (lily), which were isolated specimens in Japan, Korea and Taiwan, unable to support satellite RNA (satRNA) accumulation. In order map the CMV sequences that are involved satRNA support, HL-CMV (Japanese lily isolate), Y-CMV (ordinary strain) Y-satellite (Y-sat) used as source material. The pseudorecombinants between revealed RNA1 was essential for replication lily. results chimeric constructs various mutations showed two amino acid residues (at positions 876 891) 1a protein determinants inability a satRNA. Specifically, Thr at position had more pronounced effect than Met 891. Specific changes sequence also detected 3' terminus Y-sat these particular alterations allowed it be supported by HL-CMV. It is believed that, through evolution, adaptation resulted introduction protein, coincidentally affected ability satRNAs.
