ABSTRACT
Summary Cells of the trophoblast lineage constitute major part placental tissues in higher mammals. Recent derivation human stem cells (TSC) from cytotrophoblasts (CT) and naïve PSCs opens new opportunities for studying development function placenta. Several recent reports have suggested that retain an exclusive potential to give rise bona fide TSCs. Here we report inhibition TGFβ pathway avoiding WNT stimulation, leads direct robust conversion primed pluripotent into Systematic side by comparative analysis showed latter are equivalent previously derived TSC lines. Primed PSC lines exhibit self-renewal, able differentiate main lineages, present RNA epigenetic profiles indistinguishable placenta or PSCs. Our findings underscore a residual plasticity allows converting directly pre-implantation extra-embryonic cell lineages. Highlights readily convert TSCs upon TGF CHIR inhibits but not naive hPSCs line YAP is sufficient induction necessary maintenance.
