ABSTRACT
Oxidative stress is one of the leading factors responsible for poor post-thaw semen quality because overproduction reactive oxygen species (ROS) over neutralizing antioxidants present in semen. Mainly two ROS generation sites are spermatozoa, that is, mitochondria and plasma membrane. Therefore, idea targeting these specific minimization production with compounds having known mechanism actions was built up as a core this research.Present study done to investigate effects Mito TEMPO acetovanillone individually combination on freezability buffalo spermatozoa. For experiment, extender supplemented (50 μM), + μM+ 50 designated Group II, III, IV, respectively. Control group without any supplementation I. A total 24 ejaculates individual progressive motility (IPM) ≥70% were selected study. After final dilution, filling-sealing straws, equilibration, freezing per standard procedure. Semen samples evaluated IPM, membrane integrity, lipid peroxidation, antioxidant capacity (TAC), cholesterol phospholipids (C/P) ratio at both fresh stages. Evaluation ROS, mitochondrial potential (MMP), capacitation status (CTC assay), vitro fertility conducted only frozen-thawed samples. The addition μM) significantly (p < 0.05) improved terms TAC, content, C/P ratio, MMP, Chlortetracycline (CTC)-Full (F) pattern, zona binding ability while reduced production, like changes compared control group. As acts an SOD mimetic also detoxifies ferrous iron level, it aids neutralization excessive minimizes oxidative stress-related damages enhances sperm mitochondria. Earlier studies indicated μM group. The improvement observed might be inhibition Nicotinamide adenine dinucleotide phosphate (NADPH) oxidase enzyme activation by various physical/chemical inducers during cryopreservation process leads CatSper channel resulting calcium influx, premature capacitation, acrosomal reaction through adenylate cyclase cAMP/PKA-mediated tyrosine phosphorylation proteins. Acetovanillone prevents NADPH oxidase-mediated glutathione reductase activity, which has vital role protecting structural functional integrity membrane. Results beneficial efficient sustaining quality.
