ABSTRACT
Abstract Posterior capsular opacification ( PCO ) is the major complication arising after cataract treatment. occurs when lens epithelial cells remaining following surgery LC s) undergo a wound healing response producing mixture of α‐smooth muscle actin (α‐ SMA )‐expressing myofibroblasts and fibre cells, which impair vision. Prior investigations have proposed that integrins play central role in we found that, mouse cell removal model surgery, expression α V integrin its interacting β‐subunits β1, β5, β6, β8 are up‐regulated concomitant with α‐ s surgery. To test hypothesis functionally important pathogenesis, created mice lacking subunit all cells. Adult lenses transparent show no apparent morphological abnormalities compared control lenses. However, surgical removal, eyes increased proliferation, , β1‐integrin, fibronectin, tenascin‐C transforming growth factor beta TGF ‐β)–induced protein within 48 hrs, while exhibited much less proliferation little to up‐regulation any fibrotic markers tested. This effect appears result from known roles latent ‐β activation as null do not exhibit detectable SMAD ‐3 phosphorylation this robustly lenses, consistent for . These data suggest therapeutics antagonizing function could be used prevent
