Functional interleukin‐6 receptor‐α is located in tanycytes at the base of the third ventricle

ABSTRACT
Interleukin (IL)-6- /- mice develop mature onset obesity, whereas i.c.v. injection of IL-6 decreases obesity in rodents. Moreover, levels cerebrospinal fluid (CSF) were reported to be inversely correlated with humans. Tanycytes lining the base third ventricle (3V) hypothalamus have recently been importance for metabolism. In present study, we investigated whether tanycytes could respond CSF. With immunohistochemistry using a well characterised antibody directed against ligand binding receptor IL-6, α (IL-6Rα), it was found that tanycytes, identified by two markers, vimentin and dopamine- cAMP-regulated phosphoprotein 32 kDa, contained IL-6Rα. There fewer IL-6Rα on another type ventricle-lining cells, ependymal as marker glucose transporter-1. To demonstrate immunoreactive responsive injected This treatment increased phosphorylated signal transducer activator transcription-3 (pSTAT3) after 5 minutes cells medial part arcuate nucleus 15 minutes. Intracerebroventricular leptin exerted similar effects. As expected, i.p. also induced pSTAT3 staining hypothalamus, had little effect this parameter. or vehicle only no pSTAT3-immunoreactivity. summary, there are functional at bottom 3V, agreement possibility ventricular administration via an cell type.
