ABSTRACT
Summary Flow cytometric sorting of individual chromosomes and chromosome‐based sequencing reduces the complexity large, repetitive Triticeae genomes. We flow‐sorted chromosome 5D Aegilops tauschii , D genome donor bread wheat sequenced it by Roche 454 GS FLX platform to approximately 2.2x coverage. Repetitive sequences represent 81.09% survey this chromosome, Class I retroelements are prominent type, with a particular abundance LTR/Gypsy superfamily. Nonrepetitive were assembled cover 17.76% total regions. Up 6188 nonrepetitive gene loci predicted be encoded chromosome. The numbers chromosomal distribution patterns tRNA genes suggest in L ys M et species, while assembly reveals A la species as most abundant type. comparative analysis genomic indicates conservation content. Orthologous unique genes, matching sequences, numbered 3730 barley, 5063 Brachypodium 4872 sorghum 4209 rice. In study, we provide chromosome‐specific view into structure organization Ae. ancestor wheat. This study contributes our understanding chromosome‐level evolution presents valuable resource genomics due recent hybridization its tetraploid ancestor.
