ABSTRACT
Porcine spermatogonia stem cells (pSSCs) are the only type of somatic cell that can pass genetic information to successive generations. Little is known about pSSCs vitality in vitro, and due their increasing importance research, here, we optimized a protocol culture explored potential fate vitro. Utilizing feeder-free system with 2D peptide-coating small chemical molecules (including CHIR99021, Repsox, vitamin C, folic acid, CD lipid concentrate), were able prolong time by at least three months compared previous methods. Moreover, found could proliferate self-renew seminiferous tubules infertile mice. However, they not perform meiosis. Our study shows this optimizes may facilitate advanced research on SSC biology manipulation germ cells.
