Point-of-care applicable metabotyping using biofluid-specific electrospun MetaSAMPs directly amenable to ambient LA-REIMS

ABSTRACT
In recent years, ambient ionization mass spectrometry (AIMS) including laser ablation rapid evaporation IMS, has enabled direct biofluid metabolome analysis. AIMS procedures are, however, still hampered by both analytical, i.e., matrix effects, and practical, sample transport stability, drawbacks that impede coverage. this study, we aimed at developing biofluid-specific sampling membranes (MetaSAMPs) offer a directly applicable stabilizing substrate for AIMS. Customized rectal, salivary, urinary MetaSAMPs consisting of electrospun (nano)fibrous blended hydrophilic (polyvinylpyrrolidone polyacrylonitrile) lipophilic (polystyrene) polymers supported metabolite absorption, adsorption, desorption. Moreover, MetaSAMP demonstrated superior coverage stability compared to crude analysis was successfully validated in two pediatric cohorts (MetaBEAse, n = 234 OPERA, 101). By integrating anthropometric (patho)physiological with MetaSAMP-AIMS data, obtained substantial weight-driven predictions clinical correlations. conclusion, holds great application potential on-the-spot metabolic health stratification.
