ABSTRACT
An evaluation of broth dilution antifungal susceptibility tests was performed by determining both the micro- and macrodilution MICs amphotericin B, fluconazole, ketoconazole, 5-fluorocytosine, miconazole, itraconazole against representative species opportunistic hyphomycetes (Fusarium spp. Cladosporium [Cladophialophora] spp.) ascomycetes (Chaetomium spp.). A total 78 strains were tested, majority them twice some three times on different days. Both methods according to recommendations National Committee for Clinical Laboratory Standards (Document M27-P), with exception temperature incubation, which 25 degrees C in our case. spectrophotometric method inoculum preparation, RPMI 1640 medium buffered morpholinepropanesulfonic acid (pH 7.0), an additive drug procedure used. The obtained two read after 48, 72, 96 h incubation Fusarium 96, 120 remaining isolates. kappa test used calculate degree agreement. Considering fungal groups together, a good agreement between results observed almost all drugs at times. There no cases poor highest level (kappa index = 1) ketoconazole second-day reading. These support further microdilution as alternative reference test.
