ABSTRACT
Inflammation in asthma and other allergic diseases is characterized by excessive production of immunoglobulin E (IgE) the influx leukocytes, especially eosinophils. Interleukin 4 (IL-4) IL-5 are essential for IgE eosinophilia, respectively, produced mast cells conditions, which glucocorticoids widely used therapeutically. We assessed effect on IL-4 mRNA RBL-2H3 cell line, an analog mucosal cells. mRNAs were induced antigen that to cross-link receptor bound IgE, calcium ionophore, or ionophore with phorbol ester markedly inhibited dexamethasone. In activated ester, 10(-6) M dexamethasone reduced levels only 12.8 5.7%, those without dexamethasone, 10(-9) caused reductions 27 56%, respectively. Hydrocortisone at 10(-7) almost completely production. Dexamethasone was inhibitory even if it added after activated, provided present cultures least 1.5 h. These studies indicate expression highly sensitive glucocorticoids. The data suggest these effects may contribute clinical efficacy therapy diseases.
