ABSTRACT
ABSTRACT We developed a highly sensitive PCR method that enables the diagnosis and posttherapeutic follow-up of visceral leishmaniasis with patient blood. The assay was thoroughly optimized by successive procedural refinements to increase its sensitivity specificity. It compared in vitro cultivation as well direct examination bone marrow serology. Two hundred thirty-seven patients presenting clinical signs compatible were included study. Thirty-six diagnosed having Mediterranean (MVL). Twenty-three them, including 19 AIDS patients, monitored during after treatment over period from 2 weeks 3 years. Our proved more than cultivation, examination, serology for all patients. is simple can be adapted routine hospital diagnostic procedures. For primary MVL, versus 97 55% peripheral blood 100 81% samples. Regarding follow-up, overall, 48% positive samples detected only. Seven presented relapse study; six relapses at first only, sometimes few before reappearance or symptoms. conclude an well-mastered sample sufficient provide secure immunocompromised most immunocompetent also suggest systematic monitoring
