ABSTRACT
The peroxisome proliferator-activated receptor gamma (PPARgamma) regulates adipogenesis, lipid metabolism, and glucose homeostasis, roles have emerged for this in the pathogenesis treatment of diabetes, atherosclerosis, cancer. We report here that induction PPARgamma activator adipogenesis forced by overexpression adipogenic regulatory proteins is blocked upon expression dominant-negative BRG1 or hBRM, ATPase subunits distinct SWI/SNF chromatin-remodeling enzymes. demonstrate histone hyperacetylation binding C/EBP activators, polymerase II (Pol II), general transcription factors (GTFs) initially occurred at inducible PPARgamma2 promoter absence function. However, GTFs were subsequently lost from cells expressing SWI/SNF, explaining inhibition expression. To corroborate these data, we analyzed interactions differentiating preadipocytes. Changes structure, hyperacetylation, Pol II, most preceded interaction enzymes with promoter. gene only subsequent association TFIIH Thus, nuclear hormone during requires to facilitate preinitiation complex
