ABSTRACT
Primordial germ cell 7 (PGC7)(Dppa3 or Stella) is a small inherently disordered protein that mainly expressed in oocytes and plays vital role the regulation of DNA methylation reprogramming imprinted loci through interaction with other proteins. Most PGC7-deficient zygotes are blocked at two-cell stage an increased tri-methylation lysine 27 histone H3 (H3K27me3) level nucleus. Our previous work has indicated PGC7 interacts yin-yang1 (YY1) essential for recruitment enhancer zeste homolog 2 (EZH2)-containing Polycomb repressive complex (PRC2) to H3K27me3 modification sites. Here, we found presence weakened between YY1 PRC2 without disrupting assembly core subunits complex. In addition, promoted AKT phosphorylate serine 21 EZH2, resulting inhibition EZH2 activity dissociation from YY1, thereby decreasing level. zygotes, inhibitor MK2206 both enter pronuclei but disturbing subcellular localization caused increase pronuclei, as well expression zygote-activating genes regulated by embryos. summary, could affect zygotic genome activation during early embryonic development regulating recruitment, activity, localization.NEW & NOTEWORTHY inhibits YY1. promotes pEZH2-S21 level, which weakens interaction, promote embryos, ultimately affects embryo development.
