ABSTRACT
We have investigated the role of p55 and p75 tumor necrosis factor receptors 1 2 (TNFR1 TNFR2, respectively) in TNF-induced alteration endothelial permeability vitro vivo. Stimulation TNFR1 with an agonist antibody or a receptor-selective TNF mutein increased flux 125 I-albumin through cell monolayers. An antagonist anti-TNFR1 antibody, but not anti-TNFR2 antibodies, blocked activity vitro. TNFR1, induced cytoskeletal reorganization associated permeability. SB-203580, p38 mitogen-activated protein kinase inhibitor, TNFR1-induced A selective mouse human TNF, which binds to murine leakage trypan blue-albumin from liver vessels mice. These results indicate that stimulation is necessary sufficient increase However, anti-murine TNFR2 partially inhibited effect on vessels, suggesting also plays regulation vascular
