ABSTRACT
San-Huang-Chai-Zhu formula (SHCZF) has been used to improve cholestasis for many years. This study aims predict the possible gene targets of SHCZF in treating acute intrahepatic (AIC) rats.Eighteen SD rats were randomly assigned normal group, ANIT and + group. Alpha-naphthylisothiocyanate (ANIT) was induce AIC. Serum biochemical indexes detected each After treatment, livers collected extract RNA. The library constructed by TruSeq RNA, sequenced Illumina, analyzed various bioinformatics methods. qRT-PCR verify target genes related efficacy SHCZF.Serum ALT, AST, ALP, TBIL significantly higher group than ALT AST levels substantially lower those A total 354 intersected screened expression level correlation PCA analysis, GO KEGG pathway enrichment WGCNA STEM analysis. Then, 4 overlapping found pathway/BP/gene network construction. reversed downregulation CYP4A1 HACL1 upregulation DBI F11R induced ANIT. In addition, result showed that mRNA CYP4A1, HACL1, liver consistent with prediction analysis.CYP4A1, are occurrence ANIT-induced AIC may be considered as treatment
