ABSTRACT

Abstract
During progression from single cancer cells to a tumor mass and metastases, send signals that can subvert their tissue microenvironment. These involve soluble molecules various extracellular vesicles, including particular type termed exosomes. The specific roles of exosomes secreted in the microenvironment, however, is unclear. small GTPases RAB27A RAB27B regulate exocytosis multivesicular endosomes, which lead exosome secretion, human HeLa cells. Here, we used mouse models show Rab27a blockade mammary carcinoma decreased secretion characterized by endocytic markers, but also matrix metalloproteinase 9, not associated with resulted primary growth lung dissemination metastatic (4T1), nonmetastatic (TS/A). Local 4T1 tumors required mobilization population neutrophil immune induced Rab27a-dependent together combination cytokines and/or metalloproteinases. Our findings offer in vivo validation concept exert key pathophysiologic during formation progression, they highlight idiosyncratic character context. Cancer Res; 72(19); 4920–30. ©2012 AACR.
