ABSTRACT

Abstract
Purpose: Cancer-associated fibroblasts (CAF) are essential components of the stroma that play a critical role in cancer progression. This study aimed to identify novel CAFs markers might contribute invasion and prognosis colorectal cancer.

Experimental Design: The azoxymethane/dextran sodium sulfate mouse model sporadic colon represents an adequate source for isolation normal fibroblasts. By using explants technique, we purified from tissues. Whole-cell extracts supernatants were subjected in-depth quantitative proteomic analysis by tandem mass spectrometry. Further validations upregulated proteins carried out chemokine microarray immunohistochemical analyses human tissues.

Results: Using fold-change 1.4 or more, found 132 125 differentially expressed whole-cell supernatants, respectively. We CAFs-associated proinflammatory desmoplastic signatures. signature was composed several cytokines. Among them, CCL2 CCL8 caused increase migration KM12 cells. 30 secreted proteins. In samples, expression LTBP2, CDH11, OLFML3, and, particularly, FSTL1 significantly increased tumoral stroma, without significant epithelial combination CALU CDH11 stromal showed association with disease-free survival poor prognosis.

Conclusion: have identified as selective biomarkers candidate prognostic significance cancer. Clin Cancer Res; 19(21); 6006–19. ©2013 AACR.
