ABSTRACT
Introduction: Circulating microRNA (miRNAs) profiles are dysregulated in heart failure (HF) suggesting possible diagnostic and prognostic applications. In addition, miRNAs may yield insights into the pathophysiology of HF provide targets for therapeutic intervention. We sought to identify utility further investigate selected as potential Hypothesis: hypothesized that distinct circulating be potentially important therapy improve outcomes HF. Methods: Plasma levels were profiled from 338 well-characterized patients 208 age-matched controls without also examined expression a rat post-myocardial infarction (MI) model. Target predictive databases used miRNA targets. Results: Comparison between non-HF was carried out by multivariate analysis. Seventy five differed significantly after adjustment age, hypertension, diabetes atrial fibrillation (p-value FDR<0.01). Twenty four with AUC>0.65 , p<0.001 (for discrimination on receiver-operator analysis) highly up- or down-regulated clinical data comparing vs plasma samples post-MI Results showed miR-503 (AUC=0.73, FC (Fold Change) =1.69, p<0.001) higher miR-374b-5p (AUC=0.71, FC=-1.45, lower when controls. experimental model, (FC=2.21, p=0.017) increased (FC=-1.35, p=0.02) decreased at Day 2 7, but not significant 14 sham. MiR-503 has been reported target CCNE1 cdc25A. The predicted miR-374b is NPPB . roles warrant investigation using both in-vitro in-vivo preclinical models. Conclusion: detailed study profiling demonstrated associated
