ABSTRACT
Background The mechanisms involved in the dysfunction of both endothelium-dependent vasodilatation (EDV) and NO biosynthesis related to smoking are unclear. In this study, EDV was assessed healthy smokers nonsmokers vivo and, using serum from same individuals, biosynthetic pathway vitro. Methods Results Flow-mediated brachial artery measured 23 male patients (8 15 smokers). Serum collected, added confluent (≈85%) monolayers human umbilical vein endothelial cells (HUVECs), incubated for 12 hours. Basal substance P–stimulated production measured. HUVECs used measuring basal were lysed, synthase (eNOS) protein expression eNOS activity determined. lower compared with ( P <0.001). treated showed significantly <0.0001) stimulated <0.02) production, higher <0.0001), but <0.004). There a significant positive correlation between vitro (rho=0.57, <0.01) r =0.54, <0.008) negative =−0.60, <0.003). Conclusions This is first study combine an model near-physiological demonstrate association decreased reduced EDV. Cigarette associated EDV, generation, presence increased expression.
