Rapid Detection of DNA Methylation with a Novel Real-Time Fluorescence Recombinase-Aided Amplification Assay

ABSTRACT
Researchers have conducted in-depth research on DNA methylation mechanism, which is related to various diseases such as deficiency of imprinted gene and occurrence tumors. This study provides a novel rapid quantitative detection assay real-time fluorescence recombinase-aided amplification (RAA) for methylation. Firstly, specific sequence genes was chosen primers fluorogenic probe RAA experiment were designed synthesized. Lastly, these products proven by sequencing analysis. Results showed that the efficiency template concentration had linear dependent ( R 2 > 95%) when range 4.64×10 8 copies/ μ L˜4.64×10 4 L. The test can also detect positive samples below Remarkably, entire process only takes 15–20 minutes, so it beneficial bedside simple screening some special sites, resistance genes. In word, this method has very great potential with in clinical settings, especially if analysis needs be done quickly easily.
