Construction of Deoxynivalenol Enrichment and Detection System Based on Magnetosome Surface Display

ABSTRACT
Deoxynivalenol (DON) is a mycotoxin commonly found in cereals. It has strong toxicity with high stability, thus it easy to remain food and cause serious poisoning symptoms consumers. To establish an efficient simple DON detection system, biological nano magnetic particle called bacterial magnetosomes (BMs) extracted from magnetotactic bacteria ultrasonic crushing adsorption was used this study. A single-stranded DNA aptamer specifically binding coupled the surface of MBs by two different crosslinking agents, glutaraldehyde polyethyleneimine (PEI), respectively, synthesize kinds BMs-aptamer complexes for enriching DON. Then, rate complex determined HPLC. In results, absolute capacity 1 mg 27.24 ng when as crosslinker 27.64 PEI crosslinker. The optimization results desorption conditions showed that under optimal elution (D Nase I+methanol 2 times), adsorbed BMs-glutaraldehyde-aptamer reached 72.7%, while BMs-PEI-aptamer 64.1%. Overall, current study enriched applications detection, also provides new idea enrichment recovery
