Heavy ion mutagenesis combined with triclosan screening provides a new strategy for improving the arachidonic acid yield in Mortierella alpina

ABSTRACT
Arachidonic acid (ARA), which is a ω-6 polyunsaturated fatty acid, has wide range of biological activities and an essential component cellular membranes in some human tissues. Mortierella alpina the best strain for industrial production ARA. To increase its yield arachidonic heavy ion beam irradiation mutagenesis was carried out combination with triclosan octyl gallate treatment. The obtained mutant F-23 ultimately achieved ARA 5.26 g L− 1, 3.24 times higher than that wild-type strain. In addition, quantitative real-time PCR confirmed expression levels synthase (FAS), Δ5-desaturase, Δ6-desaturase, Δ9-desaturase were all significantly up-regulated strain, especially Δ6- Δ9-desaturase, 3- 2-fold, respectively. This study feasible breeding strategy improving provided high yield.
