ABSTRACT
IL-1β was considered as an important inflammatory cytokine in diabetic cardiovascular complications. DCM is one of the major manifestations complications whose specific mechanisms are still unclear. In this study, we investigated role myocytes apoptosis DCM. vitro high- glucose medium and/or were used to incubate isolated primary myocytes. siRNA knockdown irak2 gene expression. Apoptosis evaluated by Hoechst and TUNEL staining. vivo rats induced STZ injection confirmed cardiac hemodynamic determinations. The IL-1 receptor antagonist, IL-1Ra also treat rats. Myocardial assessed assay. both studies, expression levels GRP-78, IRAK-2 CHOP analyzed Western Blotting. ELISA employed exam content serum cell supernatants. Myocytes not identified source secretion under incubation. High incubation elevated ER- stress mediated which attenuated silencing. Dramatically increased circulating myocardial found stimulated activation ER lead apoptosis. administration IL-1Ra, however, IRAK2/CHOP without affecting fasting blood concentration. Elevated contributed promote stress- IRAK-2/CHOP pathway
