ABSTRACT
Abstract Background Intervertebral disc degeneration (IDD) is the main cause of low back pain. Patients with pain may experience significant socio-economic burdens and decreased productivity. Previous studies have shown that inflammation one causes IDD. Astragaloside IV (AS IV), a traditional Chinese medicine, has been reported to therapeutic effects on many inflammation-related diseases; however, effectiveness AS as treatment for IDD not studied. Methods Nucleus pulposus (NP) cells from patients were used experiments. Cell counting kit 8 (CCK8) was evaluate effect viability NP (NPCs). To mimic in vitro, NPCs divided into following groups: control group, interleukin 1β (IL-1β) + IL-1β group. analyse IL-1β-induced IDD, Western blotting, RT-qPCR, flow cytometry, immunofluorescence assays performed. vivo, rat model puncture-induced established. Results effectively alleviated inflammation, apoptosis, extracellular matrix NPCs. We also observed phosphorylation inhibitor kappa B-alpha (p-IκBα) cytosol, reduced nuclear translocation NF-κB p65, indicating inhibited pathway.
