ABSTRACT
Our previous study found that suppression of TRPM7 reduced neuronal death in adult rat ischemic brain injury. It was reported carvacrol blocked and attenuated injury an MCAO model. The effects on neonatal stroke remain unknown. This investigated the behavioral impairment after hypoxia-ischemia mice potential signaling pathway underlying these effects.Carvacrol inhibited current HEK293 cells over-expressing TRPM7-like hippocampal neurons a dose-dependent manner. Carvacrol (>200 μM) OGD-induced cortical neurons. 24 hours HI, protein level ipsilateral hemisphere significantly higher than contralateral hemisphere. (30 50 mg/kg) pre-treatment infarct volume HI also improved neurobehavioral outcomes. Furthermore, animals pre-treated with had fewer TUNEL-positive compared to vehicle-treated 3 days HI. increased Bcl-2/Bax p-Akt/t-Akt ratios decreased cleaved caspase-3 expression HI.Carvacrol protects against hypoxic-ischemic by reducing volume, promoting pro-survival inhibiting pro-apoptotic signaling, as well improving neuroprotective effect may be mediated inhibition channel function. is drug development target for treatment stroke.
