ABSTRACT
Despite the overwhelming number of investigations on AKT, little is known about lncRNA AKT regulation, especially in GBM cells. RNA-binding protein immunoprecipitation assay (RIP) and RNA pulldown were used to confirm binding LINC00470 fused sarcoma (FUS). Confocal imaging, co-immunoprecipitation (Co-IP) GST assays detect interaction between FUS AKT. EdU assay, CCK-8 intracranial xenograft performed demonstrate effect malignant phenotype RT-qPCR Western blotting test pAKT. In this study, we demonstrated that was a positive regulator for activation GBM. bound form ternary complex, anchoring cytoplasm increase activity. Higher pAKT activated by inhibited ubiquitination HK1, which affected glycolysis, cell autophagy. Furthermore, higher expression associated with tumorigenesis poor patient prognosis. Our findings revealed noncanonical signaling pathway, i.e., directly interacts FUS, serving as an activator promote progression. has important referential significance evaluate prognosis patients.
