ABSTRACT
Many viral proteins are related to suppressing apoptosis in target cells and hence beneficial replication. The V protein of Newcastle disease virus (NDV) is one such that plays an important role inhibiting a species-specific manner. However, date, there have been no reports clarifying the antiapoptotic mechanisms protein. present study was undertaken determine apoptotic potential chicken embryo fibroblast cell line (DF-1 cell) elucidate its molecular action. Here, yeast two-hybrid system used screen host interact with identified thioredoxin-like 1 (TXNL1) as binding partner. Immuno-colocalization TXNL1 DF-1 further verified interaction two proteins. Through overexpression knockdown cells, effects NDV replication were examined. Cell detected by flow cytometry. mRNA expression levels Bax, Bcl-2 Caspase-3 quantitative real-time PCR (Q-PCR) Western blotting. Q-PCR plaque assay. results revealed induced inhibited cells. Furthermore, blot suggested through pathway involving Bcl-2\Bax Caspase-3. Finally, this work provides insight into mechanism which inhibits apoptosis.
