ABSTRACT
The egg production performance of chickens is affected by many factors, including genetics, nutrition and environmental conditions. These factors all play a role in affecting the development follicles. MicroRNAs (miRNAs) are important non-coding RNAs that regulate biological processes targeting genes or other after transcription. In animal reproduction process, miRNA known to affect atresia follicles regulating apoptosis autophagy granulosa cells (GCs).In this study, we identified potential miRNAs atretic broody unatretic healthy chickens. We gga-miR-30a-5p 50 differentially expressed found played regulatory chicken function miR-30a-5p was explored through transfection test inhibitor mimics. used qPCR, western blot flow cytometry detect cell apoptosis, steroid hormone synthesis. Confocal microscopy transmission electron for observation autophagolysosomes. levels estradiol (E2), progesterone (P4), malondialdehyde (MDA) superoxide dismutase (SOD) were detected ELISA. results showed negative effect on cells, also contributed hormones reactive oxygen species (ROS) production. addition, obtained from biosynthesis dual luciferase experiments Beclin1 target gene miR-30a-5p. rescue experiment conducted further confirmed belongs pathway.In summary, deep sequencing follicles, indicated inhibits death inhibiting Beclin1.
