ABSTRACT
This study compares the effect of nanosized silicon dioxide (nanosized SiO 2 ) and microsized (microsized particles on fibrogenesis in rats. Wistar rats were instilled intratracheally with saline, 20 mg or , sacrificed at 1 months after instillation. The lungs analysed for changes lung/body coefficient hydroxyproline content. Changes pathology fibrotic grade observed by use hematoxylin eosin Van Gieson dyeing. expression interleukin-4 (IL-4) transforming growth factor-b1 (TGF-b1) was immunohistochemical technique, protein quantitatively image analysis. content groups significantly lower than those both instillation (PB / 0.05 PB 0.01), but without significant differences from saline control groups. At month instillation, there mainly cellular nodules (Stage I) group, while group Stage II, II silicotic observed. still I group. In III found. Quantity analysis showed that expressions IL-4 TGF-b1 difference Our experiment revealed might be milder rats, potentially resulting nanoparticals tending to diffused easily translocated due their ultrafine particle size compared particles.
