ABSTRACT
We generated a Spot 14 null mouse to assess the role of in de novo lipid synthesis and report exhibits phenotype lactating mammary gland. pups nursed by dams gain significantly less weight than wild-type dams. In contrast, heterozygous show similar littermates. found triglyceride content milk is reduced. demonstrate this reduction direct result decreased glands, corroborated marked medium-chain fatty acids pool. Importantly, reduced lipogenic rate not associated with significant changes activities or mRNA key enzymes. Finally, we expression 14-related gene liver adipose tissue, which absent suggest that both proteins required for maximum efficiency vivo these impart novel mechanism regulating lipogenesis.
