ABSTRACT
ABSTRACT The Golgi complex is responsible for processing and sorting of secretory cargos. Microtubules are known to accelerate the transport proteins from endoplasmic reticulum (ER) plasma membrane. However, whether post-Golgi strictly requires microtubules still unclear. Using retention using selective hooks (RUSH) system synchronize trafficking cargos, we show that anterograde tumor necrosis factor (TNF) strongly reduced without microtubules. We two populations elements co-exist in these cells. A centrally located giantin-positive sustains trafficking, newly formed peripheral mini-stacks accumulate cargos cells a genome-edited GFP–giantin cell line, observe trafficking-competent population corresponds pre-existing was present before removal All support after long-term depletion relocation ER treatment with Brefeldin A. Our results demonstrate functional maturation needed ensure microtubule-driven not required.
