ABSTRACT
In this study, we develop methods to measure galvanotaxis of fibroblasts and determined the optimum conditions electrical stimulation.An inverted 35mm dish containing cell suspensions (3×105 primary human skin fibroblasts, DMEM, 10% FBS) was placed on centre a 100mm dish. The removed 24 hours later, culture medium added Fibroblasts were randomised (double-blind) into three groups, where stimulation given at varying intensities: 0UA (control), 50UA, 100UA. Electrical (frequency=0.3Hz) conducted, for duration 4 hours, with platinum electrodes in CO2 incubator. We took pictures immediately before 20 after stimulation. calculated migration ratio negative pole by dividing area attached that stimulation.The significantly higher 100UA group than control (p<0.05). ratios 0.902±0.292 group, 1.128±0.253 50UA 1.24±0.300 group.This study observed change proliferation during initial 24-hour period plating thus able quantitatively evaluate migration. results suggest low-intensity direct current promotes dermal which is charged positive electricity. Several clinical reports using showed microcurrent efficacy pressure ulcer healing. based our vitro experiment might be important development physical therapy ulcers.
