ABSTRACT
Abstract A number of early biochemical responses bone cells to mechanical loading have been identified, but the full sequence events from sensing strain formation new is poorly characterized. Extracellular matrix proteins can modulate cell behavior and would be ideal molecules amplify response loading. The extracellular protein, tenascin-C, supports differentiation cultured osteoblast-like cells. current study was carried out investigate whether expression patterns tenascin-C in loaded bones support a role for this protein as mediator osteoregulatory Tenascin-C investigated by Northern blot analysis rat ulnae subjected an established noninvasive regimen engendering physiological levels. RNA extracted compared with contralateral control 6 h after showed significant increase transcript expression. presence immunohistochemistry animals killed 3, 5, or 15 days initiation daily In at 3 5 days, periosteal surfaces undergoing load-induced reversal resorption enhanced staining. formed clearly demarcated old strong staining lines. Within bone, seen lacunae older not more recently embedded osteocytes. results presented here indicate that osteogenic This may acts mechanically adaptiveresponse.
