ABSTRACT
Common thymic epithelial progenitor/stem cells (TEPCs) differentiate into cortical and medullary (TECs), which are required for the development selection of thymocytes.Mature TEC lines have been widely established.However, establishment TEPC is rarely reported.Here we describe stomal cell lines, named TSCs, from fetal thymus.TSCs express some markers present on tissue such as Sca-1.Gene expression profiling verifies identity TSCs.RANK stimulation these induces autoimmune regulator (Aire) Aire-dependent tissue-restricted antigens (TRAs) in TSCs vitro.TSCs could be differentiated cell-like with exogenously expressed NF-κB subunits RelB p52.Importantly, upon transplantation under kidney capsules nude mice, able to mature TEC-like that can support limited T vivo.These findings suggest TSC established bear characteristics functional vitro vivo.The cloned TEPC-like may provide useful tools study differentiation precursors.
