ABSTRACT
A synergistic combination of two next-generation sequencing platforms with a detailed comparative BAC physical contig map provided cost-effective assembly the genome sequence domestic turkey (Meleagris gallopavo). Heterozygosity sequenced source allowed discovery more than 600,000 high quality single nucleotide variants. Despite this heterozygosity, current (∼1.1 Gb) includes 917 Mb assigned to specific chromosomes. Annotation identified nearly 16,000 genes, 15,093 recognized as protein coding and 611 non-coding RNA genes. Comparative analysis turkey, chicken, zebra finch genomes, comparing avian mammalian species, supports characteristic stability genomes identifies genes unique lineage. Clear differences are seen in number variety immune system where expansions novel less frequent examples gene loss. The provides resources further understand evolution vertebrate genetic variation underlying economically important quantitative traits poultry. This integrated approach may be model for providing both chromosome level assemblies other species agricultural, ecological, evolutionary interest.
