ABSTRACT
During muscle development, myosin and actin containing filaments assemble into the highly organized sarcomeric structure critical for function. Although sarcomerogenesis clearly involves de novo formation of filaments, this process remained poorly understood. Here we show that mouse Drosophila members DAAM formin family are sarcomere-associated assembly factors enriched at Z-disc M-band. Analysis dDAAM mutants revealed a pivotal role in myofibrillogenesis larval somatic muscles, indirect flight muscles heart. We found loss function results multiple defects sarcomere development including thin thick filament disorganization, M-band formation, near complete absence myofibrillar lattice. Collectively, our data suggest is required initial subsequently it promotes elongation by assembling short polymers anneal to pointed end growing antagonizing capping protein Tropomodulin.
